چکیده

As one common urinary/reproductive system malignant tumor in males, the incidence of prostate cancer is increasing globally. Hepsin is one transmembrane protein and exerts critical function for cellular integrity. Previous study has suggested the pivotal role of Hepsin in infiltration and invasion of tumor cells. This study thus investigated the expression profile of Hepsin in prostate cancer and its biological significance. A total of 40 prostate cancer patients were from January 2013 to December 2014 in our hospital were recruited in this study, in parallel with 10 cases of benign prostatic hyperplasia (BPH) patients. Tissue samples were collected during surgery. In situ hybridization (ISH) and RT-PCR were performed to detect Hepsin expression level. RNA interference (RNAi) technique was employed to inhibit the expression of Hepsin in prostate cancer cell line 22RV1. MTT assay was further used to evaluate cell proliferation while Transwell approach was applied for cell invasiveness. Hepsin expression was significantly elevated in prostate cancer tissues compared to BPH tissues (P<0.05). With advanced stage of cancer, Hepsin expression level was further elevated. When Hepsin expression was knocked-down in 22RV1 cells, the invasion ability was decreased by 45.5% but not for cell proliferation. The expression level of Hepsin had no significant effects on prostate cancer cell proliferation but significantly modulated migration and invasion of tumor cells.

HEPSIN inhibits cell growth/invasion in prostate cancer cells.

Expression of HEPSIN, a type II transmembrane serine protease in prostate cancer (CaP), has been highlighted by several studies analyzing CaP-specific gene expression alterations by cDNA microarray. Evaluations of the biological functions of HEPSIN in CaP cells are warranted for better assessment of its utility as a biomarker and/or therapeutic target. In stable clones of PC-3/HEPSIN transfecta...

The IGSF1, Wnt5a, FGF14, and ITPR1 Gene Expression and Prognosis Hallmark of Prostate Cancer

Background: Prostate cancer is considered as the second leading cause of cancer related death in men worldwide and the third frequent cancer among Iranian men. Despite the use of PSA as the only biomarker for early diagnosis of prostate cancer, its application in clinical settings is under debate. Therefore, the introduction of new molecular markers for early detection of prostate cancer is nee...

Hepsin is highly over expressed in and a new candidate for a prognostic indicator in prostate cancer.

PURPOSE Other cDNA microarray studies have shown that hepsin is one of the highly over expressed genes in prostate cancer tissue compared with nonmalignant and benign prostatic hyperplasia tissue. We quantitatively analyzed hepsin gene expression with real-time polymerase chain reaction and calculated its relationships with clinicopathological parameters in a large cohort of samples. Pegylated kunitz domain inhibitor suppresses hepsin-mediated invasive tumor growth and metastasis.

The transmembrane serine protease hepsin is one of the most highly upregulated genes in prostate cancer. Here, we investigated its tumor-promoting activity by use of a mouse orthotopic prostate cancer model. First, we compared the tumor growth of low hepsin-expressing LnCaP-17 cells with hepsin-overexpressing LnCaP-34 cells. After implantation of cells into the left anterior prostate lobe, LnCa...

Study of NGEP expression in androgen sensitive prostate cancer cells: A potential target for immunotherapy

Background: Prostate cancer is one of the leading causes of cancer deaths among men. New gene expressed in prostate (NGEP), is a prostate-specific gene expressed only in normal prostate and prostate cancer tissue. Because of its selective expression in prostate cancer cell surface, NGEP is a potential immunotherapeutic target. To target the NGEP in prostate cancer, it is essential to investig...
